He played in a game against his former Cowboys team and finished with 110 receiving yards and a touchdown. He inked the dotted line on July 29, 2020, and became an active/non-football injury player on August 12, 2020.

In 2020, Cole led the Bills to a winning record. It was in the wild card round when he played for the Buffalo Bills against the Indianapolis Colts, catching seven passes for 57 yards. In the game against the Los Angeles Rams, he hauled in six passes.

In a game against the New York Jets, he had 11 receptions. Against the Arizona Cardinals, he tallied 11 catches. With seven receptions, Cole set a new postseason record for most catches in a game.
